5.In this experiment, the 3′-end of the guide strand RNA is labeled by 6-FAM. The 3′-end of the single-stranded RNA is anchored within the nucleic acid binding pocket of the PAZ domain in the Argonaute-guide RNA binary complex [5, 11–18]. However, the deletion of the interaction between the 3′-end of the guide strand and the PAZ domain does not seem to have an effect on the binding affinity of the Argonaute-guide binary complex [19, 20].
